The German edged to a serve-dominated 7-6 7-6 win against the American to tee up a clash with Mattia Bellucci.
The Italian progressed after upsetting seventh seed Alejandro Davidovich Fokina by a 6-2 6-7 6-1 scoreline.
This will be their maiden meeting on tour although Hanfmann did win a narrow qualifier 7-6 6-4 at the French Open three years ago.
The 34-year-old German has the more powerful serve and at 59 in the world he is 19 places better on the ATP ladder.
That said, Bellucci is a real threat when his game clicks and there’s scope for this to have plenty of holds and lots of games.
